The fantasy world of molecular machines reveals itself before the eyes of both scientists: channels, barriers and valves - elements necessary for cells to their functions.


    The discovery by the two scientists explains how salt (ions) and water are transported outside and inside the cells of the body. They give as an example the way kidneys recover water from primary urine and how electrical signals in our nerve cells are generated and disseminated. This is extremely important for the study of many diseases related to kidney, heart, muscles and nervous system.


    Water goes through the cell walls, even only for maintaining the cell pressure. The appearance and function of the "pores" of the cell membrane through which water molecules pass, remain one of the classic unsolved matters of biochemistry for long time . That is until 1990 when Peter Agre discovered the first water channel. He called them aquapores.


    Water molecules are not the only units passing in and out the cell. There is a need of coordination, "communication" between the cells, so they could be able to function as something more than a biological mass. The signals sent to and between cells consist of ions. For example, the brain signals include such chemical reactions. Information is passed from cell to cell via chain of nerve cells that interact with chemical signals and ion flow.


    In 1998 Roderick MacKinnon succeeded for the first time to show how ion channels in the cell membrane looked like- an achievement which, together with that of water channels (aquapores) reveals completely new areas in biochemistry and biology.

    What are energy channels, called by the traditional Chinese medicine - energy meridians?

    WATER CHANNELS

    In the middle of the XIX century it had already been clear that there must be pores in the cell membrane, allowing water and salt streams to move. In mid-1950 years, it became clear that water moves rapidly within and outside the cell through pores that admit water molecules only. Over the next 30 years the process was studied in details and reached the conclusion that there must be some sort of a selective filter that prevents ions to pass through the cell membrane, while the neutral water molecules pass freely. Thousands of millions of water molecules pass in a single channel per second. Although it was known until 1992 nobody has been able to explain how this molecular mechanism looks, i.e. to identify which protein or proteins form the actual channel.

    In mid-1980s Peter Agre explored different membrane proteins of red blood cells. One of them he found in the kidneys. Having defined the peptide and the DNA sequence, he realizes that it must be the protein, which many have seen before – a cell water channel.

    What is osmosis?

    The liquid pressure in the cells of plants and animals is maintained through osmosis. In this process small molecules (like water) pass through a semi-permeable membrane. If the membrane does not let macromolecules or salts, that are in higher concentration, to pass on the other side, then small molecules (water) will pass on that side, trying to "distribute" substances that can not pass through the membrane. The osmolality is usually the reason for the cells to bloate and become hard, as in a flower stalk, for example.

    Peter Agre also knew that mercury ions and heavy metals prevent all cells from absorbing and releasing water, and an experiment showed that mercury ion cells behave the same way with the protein cells he found. This made him believe his search for the protein was over . He named it „aquapore”.


    How does the water channel work? Form and Function

    In 2000, Agre and his colleagues presented the first pictures of a 3D aquapore structure. It became possible to trace in details how water channels work with this information. How did they let only water molecules, but do not let other molecules or ions? Membranes, for example, do not allow protons (elementary particles with positive electric charge) to pass. This is a very important fact, since the difference in concentration of protons inside and outside the cell is a cell based system for energy storage.

    Selectivity is a fundamental characteristic of the channel. Water molecules squeeze through a narrow channel, directed by the local electric field, formed by the atoms of the channel wall. The protons (or H3O+ions) are blocked on their way and rejected because of their positive charge

    Medical importance of the water channels.

    Over the past 10 years, water channels are subject to extensive research. The aquapores turned out to be one big family. Such as in bacteria, plants and animals. Until now only in the human body were discovered at least 11 different variants of aquapores. The function of these proteins is studied in details with a focus on their role in the physiological processes. In the human body the water channels play an important role, especially for the functioning of the kidneys.

    KIDNEYS - ONE MIRACLE OF THE INVENTIVE NATURE

    Our kidneys are two amazing organs, which serve us in various ways. They regulate our blood pressure and are engaged in the production of vitamin D. But their most important function is to dispose of the toxic waste from the body and keep all the valuable components and blood proteins in the body. Healthy kidneys perform this important activity, using millions of filters called glomerulus. Each glomerulus is a microscopic sphere of capillaries (tiny blood vessels ) through which blood is filtered with incredibly high selectivity for water, ions and other small molecules. This fantastic selectivity is achieved thanks to the inner porous shell, with which the capillaries are coated, and is called the basal membrane.

    About 1.3 liters of blood is filtered by the human kidneys every minute, and every day - about 180 liters. All this liquid is reabsorbed through a series of mechanisms so that one day only 1 to 1.5 liters of urine is released outside. Primary urine goes from the glomerulus to a spiral tube, where about 70% of its water is reabsorbed by the AQP1 aquapore. At the end of the tube, another 10% of its water is reabsorbed through such an aquapore, AQP2. Sodium, potassium and chloride ions are absorbed in a similar way. The antidiuretic hormone (vasopressin) stimulates the transport of AQP2 to the cell membranes of cells from the tube wall and thus increases the reabsorption of water from urine.

    Agre examined the hypothesis in a simple experiment (Fig. 2) where he compared cells which contain protein and those that do not contain it. When cells are placed in aqueous solution, those which contain the protein absorb the water from the protein solution by osmosis and inflate, while the other cells which do not have this protein do not respond at all. Agre conduct studies with artificial liposome cells that are like a bubble surrounded by water and filled with water. Liposomes became permeable for water, if this protein is placed in their cell membrane.

    ION CHANNELS

    Cellular signals with salt! The first physical chemist, the German Wilhelm Ostval (Nobel Prize in Chemistry in 1909) suggests in 1890 that the electrical signals measured in a living tissue can be caused by moving ions inwards and outwards through the cell membranes. This electro-chemical idea became popular. The idea of the existence of something like narrow ion channels appears for the first time in 1920. Two British scientists Alan Hodkin and Andrew Huxley made a major breakthrough in this direction in the early 1950s and received the Nobel Prize in Physiology and Medicine in 1963. They showed how an ion transported a signal through the membrane of a nerve cell and was transmitted from nerve cell to nerve cell. The basis of these reactions involved mainly sodium and potassium ions Na+ and K+.

    So about 50 years ago, people already knew about the central functions of the ion channels. They must also have selectivity - to allow only one type of ions to pass on the other side and repel others. Also, this channel should be able to "open" and "close" and sometimes to conduct ion in one direction only. How this molecule mechanism work remained a mystery for a long time.


    Selectivity of the ion channels

    In 1970 it was known that ion channels are able to pass only a certain type of ions because they have something like an "ion filter". Of particular interest to scientists was to discover why some channels allow only potassium but not sodium ions to pass through. Scientists suggest that oxygen atoms in the protein play an important role as "substitutes" of water molecules that surround the potassium ions in aqueous solution and they should be exempted in entering into the channel.

    Scientists found it hard to prove this hypothesis because it has been difficult to determine the structure of membrane proteins and that of ion channels with an X-ray crystallography.

    Help came unexpectedly. The biochemist Roderick MacKinnon turnd to medicine and became a doctor. After working as a general practitioner for several years, he was attracted by the ion channels and began a research in this area. Realizing that better and higher resolution structures were needed to understand how ion channels work, he began to learn the basics of X-ray crystallography. Only a few years after that he amazed the scientific community by presenting a structure of an ion channel. This happened in April 1998.

    In 1998 the first accurate high-resolution structure of an ion channel was presented, called KcsA, from the bacterium Streptomyces lividans - MacKinnon revealed for the first time how the ion channel works at an atomic level. Now the ion filter, which passes potassium ions and stops sodium ions could be studied better. It is now possible not only to explain how ions pass through the channels but also could be seen in the crystal structure - surrounded by water molecules just before they lead the ion into the channel and how they found water on the other side of the filter (Figure 4)

    MacKinnon explains why potassium ions but not sodium ions are allowed through the filter: The reason is simple - because the distance between potassium ions and oxygen atoms in the filter is the same as that between potassium ions and oxygen atoms of water molecules surrounding it when it is outside the filter . So it can "slide" through the filter without a problem. Thus, the sodium ions, although smaller than the potassium ones may not pass through the channel. Sodium ions simply do not "fit" the oxygen atoms in the filter and therefore remain in the aqueous solution. The ability of channels to "strip" potassium ions from the water molecules and to leave them pass – does not require any energy - and is a kind of selective ion transport catalyst.

    A cell should be able to control the opening and closing of the ion channels. MacKinnon shows that this is achieved through a door at the bottom of the channel that opens and closes by a molecular "sensor". This sensor is located near the door. Sensors of ion channels respond to different signals - eg. increasing the concentration of calcium ions, the voltage of the cell membrane, or by connecting a signal molecule of some kind. By connecting different sensors to different ion channels, the nature has created channels that respond to many different signals.
